Abstract: Coronavirus disease 2019 (COVID-19), caused by SARS-CoV-2 (severe acute respiratory syndrome coronavirus 2) is a highly contagious disease. To tame the continuously raging outbreak of COVID-19, developing a cheap, rapid and sensitive testing assay is absolutely imperative. Herein, we developed a one-tube colorimetric RT-LAMP assay for the visual detection of SARS-CoV-2 RNA. The assay integrates Si–OH magnetic beads (MBs)-based fast RNA extraction and rapid isothermal amplification in a single tube, thus bypassing the RNA elution step and directly amplifying on-beads RNA molecules with the visualized results. This one-tube assay has a limit of detection (LOD) as low as 200 copies/mL for sample input volumes of up to 600 μL, and can be performed in less than 1 h from sample collection to result readout. This assay demonstrated a 100% concordance with the gold standard test RT-qPCR test by using 29 clinical specimens and showed high specificity. This one-tube colorimetric RT-LAMP assay can serve as an alternative platform for a rapid and sensitive diagnostic test for COVID-19 and is particularly suitable for use at community clinics or township hospitals.

摘要：冠狀病毒?。–OVID-19）是由SARS-CoV-2（嚴重急性呼吸綜合征冠狀病毒2型）引起的一種高度傳染性疾病。為了遏制持續(xù)肆虐的COVID-19病毒爆發(fā)，開發(fā)一種廉價、快速、靈敏的檢測方法勢在必行。在此，我們開發(fā)了一種單管比色RT-LAMP檢測SARS-CoV-2rna的方法。該方法將基于硅-羥基磁珠（MBs）的快速RNA提取和快速等溫擴增集成在一個試管中，從而繞過RNA洗脫步驟，直接在珠狀RNA分子上擴增，結(jié)果可視化。這種單管分析的檢測限（LOD）低至200拷貝/mL，樣品輸入量高達600μ五十、 從樣品采集到結(jié)果讀出不到1小時。用29例臨床標本進行RT-qPCR檢測，結(jié)果與金標準試驗符合率為100%，具有較高的特異性。這種單管比色RT-LAMP檢測方法可以作為COVID-19快速敏感診斷試驗的替代平臺，特別適合在社區(qū)診所或鄉(xiāng)鎮(zhèn)醫(yī)院使用。
